Jeez, E3 2012 wasn't even 5 months ago ! But If I had to say, Id want:

Info on Zelda U. A trailer or some artwork would be enough.
At least a mention of the new Smash Bros. game
A new exclusive IP for the Wii U. 3rd party or not, I don't really care
A new Pokemon Stadium/ Snap game. Or a game where you actually battle as the pokemon

Paper Mario U problably wont see the light of day this early due to the 3DS game.
And Rockstar Games problably aint that eager to put GTA V on Wii U. Either.

My hopes
- An announcement of Retro Studios new game that "Everybody wants them to work on". Damn teasers.
- Bayonetta 2 Gameplay
- All new announcements of new IPs, multiplatform-games and exclusive first- and third party titles.
- Platinum Games acquired by Nintendo
- Saints Row 4 for WiiU
- Support from Telltale Games
